چکیده
Abstract Sporadic occurrence of congenital portosystemic shunt (PSS) at a rate ∼1 out 10 among C57BL/6 mice, which are widely used in biomedical research, results aberrancies serologic, metabolic, and physiologic parameters. Therefore, mice with PSS should be identified as outliers research. Accordingly, we sought methods to, reliably efficiently, identify mice. Serum total bile acids ≥ 40 µM is bona fide biomarker but utility this limited by its cost invasiveness, particularly if large numbers to screened. This led us investigate assay urine might serve simple, inexpensive, non-invasive means diagnosis. Metabolome profiling uncovered that Krebs cycle intermediates, i.e., citrate, α-ketoglutarate, fumarate, were strikingly distinctly elevated the We leveraged iron-chelating pH-lowering properties such metabolites basis for three urine-based screening tests: urinary iron-chelation assay, pH strip test, phenol red assay. Our findings demonstrate feasibility using these colorimetric assays, whereby their readout can assessed direct observation, diagnose an rapid, manner. Application our protocols aid research enabling stratification which, present, likely confound numerous ongoing studies.

Introduction. Congenital portosystemic venous malformations are rare abnormalities in which the portal blood drains into a systemic vein and which are characterized by extreme clinical variability. Case Presentations. The authors present two case reports of a congenital extrahepatic portosystemic shunt (Type II). In the first patient, apparently nonspecific symptoms, such as headache and fatigu...
متن کاملAsymptomatic Congenital Intrahepatic Portosystemic Shunt
Congenital (spontaneous) intrahepatic portosystemic shunt is rare in the English literature. Most cases of portosystemic shunt occur after trauma, surgery, liver biopsy or as a result of chronic portal hypertension. Chronic shunting may result in encephalopathy, bleeding or hyperinsulinism. We report a case of an asymptomatic adult female with a presumed congenital intrahepatic portosystemic sh...

Liver disease is a major cause of morbidity and mortality. One of the most significant complications in patients with liver disease is the development of neurological disturbances, termed hepatic encephalopathy. The pathogenesis of hepatic encephalopathy is incompletely understood, which has resulted in the development of a wide range of experimental models. Congenital portosystemic shunt is on...

A 7-year-old boy presented with recurrent cholangitis, failure to thrive, and progressive splenomegaly. On examination, he had severe pallor, clubbing, and splenohepatomegaly. Lab results showed anemia (hemoglobin 7.2 g/dL) and normal liver functions. Abdominal ultrasound showed hepatosplenomegaly with coarsened liver echotexture and multifocal intrahepatic biliary radical (IHBR) dilatation. Th...
